LEICA SP5 Spectral 2 Photon Confocal Microscope

This expansion space houses the new state-of-the-art, high-speed 2 photon confocal microscope and is designed for live cell imaging.
1. Leica SP5 spectral 2-photon confocal microscope.

Resonant galvanometer laser scanning confocal optimized for live cell imaging. Turnkey, full control via software.

1) Leica DMI6000 (inverted) microscope with conventional fluorescence (100W Hg) and DIC optics; 10x dry , 20x, 40x, 63x water objectives, 63x oil objective.

2) 3 chilled PMT fluorescence detectors (digital spectral definition in 1 nm increments) plus one transmitted light detector. 8-, 12-, or 16-bit output.

3) 2 external APD detectors; 2 internal APD detectors. (green/red and CFP/YFP filters)

4) Spectra Physics Mai Tai broadband Ti:sapphire laser with 10W pump (tunable from 710 to 990 nm) (up to 2W output at 840 nm)

5) 3 visible lasers, 7 laser lines. 458, 496 488, 496, 514 nm Ar, 561 (10mW) diode, and 633 nm (10mW) HeNe

6) Dual scanning galvanometer mirrors (max 16000 Hz scan rate) 25 images/sec at 512x512; strip scans to 333 fps.

7) "SuperZ" galvo focusing stage, ±1.5 mm range, "instantaneous" XZY scanning.

8) AOTF attenuation of individual visible laser lines, EOM and ND filter attenuation of IR

9) Excitation and emission scanning capability with Linear unmixing

10) Automatically optimized confocal pinhole apertures

11) Digital zoom to 40X

12) Transmitted detector with LWD and oil lenses suited for SHG imaging

13) FRET, deconvolution, and 3D software options

 

2) Olympus DSU spinning Disk confocal microscope with DIC and back-thinned EMCCD camera.

An easy-to-operate motorized inverted microscope optimized for live cell confocal imaging. Interchangeable pinhole disks to optimize confocality; Hg arc lamp excitation and UV capable.

1. 10x uplapo, NA0.4 water, 20x uapo340 NA0.75 water, 40x uapo340 NA 1.15 water, 60x uplsapo NA 1.2 water, 100x plapo TIRFM NA 1.45 oil, 10x uplfln NA0.3 air objectives

2. filters for DAPI, FITC, rhodamine, CY5, CFP, GFP, YFP, DsRed (DSU and visual turrets)

3. Ludl 10-position excitation and emission filter changers

4. Olympus excitation ND changer

5. Pinhole disks optimized for 10x, 60x, and 100x

6. DIC hardware

7. Hamamatsu C9100 back-thinned EM-CCD camera (14-bits, >90%QE)

8. Slidebook 4.1 capture software, ratio/FRET module

9. 4.3GHz P4 PC, 2GB RAM, 330GB SATA drives, CDRW, DVDRW, dual LCD monitor PC, 256MB RX850 3D graphics board.
